Аннотация: На сьогоднішній день в світі спостерігається тенденція до зміни ставлення до сім'ї та сімейних цінностей. У розвинених європейських країнах спостерігається тенденція до високої сексуальної активності у чоловіків літнього віку і пізнього створення сім'ї з дітьми, що має певні труднощі у зв'язку зі зниженням вироблення тестостерону в літньому віці. Метою дослідження було встановити мікроскопічну організацію інтерстиціальних ендокріноцітов і сустентоцітов щурів, визначити джерела продукції оксиду азоту і інтенсивність окисного стресу в яєчках з експериментальної центральної депривації синтезу тестостерону з дифереліном на 90-ту добу експерименту. Експеримент був проведений на 20 зрілих білих щурах-самцях. Щури були розділені на 2 групи: контрольна група (10), експериментальна група (10), яким підшкірно вводили диферелін (трипторелін ембонат) в дозі 0,3 мг / кг активної речовини протягом 90 днів. Тривала центральна депривація синтезу тестостерону у тварин призводить до появи структурних ознак функціонального стресу в популяції сустентоцітов і інтерстиціальних ендокріноцітов, які спрямовані на підтримку секреції яєчка. Центральна депривація синтезу тестостерону протягом 90 днів викликає розвиток окисного стресу за рахунок гіперпродукції активних форм кисню і накопичення нітритів в тканини яєчка через підвищену індуцібельной активності NO-синтази.

Аннотация: Тривала центральна депривація синтезу тестостерону може призвести до біохімічним і морфологічних змін в яєчках. Вплив тривалого дефіциту тестостерону на вироблення активних форм азоту і кисню, морфологічні зміни в інтерстиційних ендокріноцитів та сустентоцитів ще повністю не вивчені. Кількість інтерстиційних ендокріноцитів знижена в порівнянні з контрольною групою, між звивистими канальцями є інтерстиційні простори з повною відсутністю інтерстиційних ендокріноцитів. Інтерстиційні ендокриноцити зменшені в розмірах, їх ядра гетерохромні. При вивченні структурної організації сустентоцитів щурів експериментальної групи в порівнянні з контрольною групою, ми виявили, що гіперплазія елементів гладкої ендоплазматичної сітки спостерігалася в цитоплазмі сустентоцітов. Кількість мітохондрій в цитоплазмі сустентоцитів зменшилася, електронна щільність мітохондріального матриксу була знижена, білкові структури були присутні або всередині вакуолей, або незалежно розташовувалися в цитоплазмі клітини. Біохімічні дослідження виявили збільшення продукції NO з індуцібельною NO-синтазою і розвиток окисного стресу. Експериментальна центральна депривація синтезу тестостерону з Дифереліном на 180-й день експерименту призводить до зрушення синтезу NO з конститутивних NO-синтази до індуцібельної NO-синтази і посилення окисного стресу за рахунок збільшення продукції супероксид-аніон-радикалів і зниження антиоксидантного захисту.

Аннотация: Порушення виробництва оксиду азоту (NO) може призводити до різних змін в різних органах і системах. Певні клінічні ситуації вимагають тривалого використання інгібіторів синтезу тестостерону. У науковій літературі є обмежена інформація про вплив тривалого позбавлення синтезу тестостерону на продукцію NO і мікроскопічну організацію сім'яників щурів. Тривала центральна депривація синтезу тестостерону призводить до ендотеліальної дисфункції, розвитку фіброзу, знижує вироблення оксиду азоту та зрушує про- / антиоксидантний баланс на користь прооксидантів без збільшення інтенсивності перекисного окислення ліпідів. Центральна депривація синтезу тестостерону призводить до фіброзу з подальшим порушенням структурної організації звивистих сім'яних канальців, порушень гемодинаміки, ендотеліальної дисфункції, збільшення щільності судинної стінки кровоносних судин і системному застою. Аннотация: Останнім часом тема професійної орієнтації для старшокласників набуває надзвичайної актуальності, оскільки поряд із вже існуючими факторами утруднення визначення професійної належності населення з’являються й інші, серед яких, зокрема, поява великої кількості нових професій, призначення яких не завжди є зрозумілим для пересічного громадянина, підвищення жорсткості вимог до претендентів на певну посаду, що спричинене досить жорсткою конкуренцією. Профорієнтаційні проекти дають змогу більш детально інформувати підростаюче покоління щодо особливостей якісної підготовки висококваліфікованих спеціалістів-медиків, а саме їх навчанням у вищому медичному закладі. Такий підхід до вибору роду діяльності викликає у школярів зацікавленість, спонукає до свідомого вибору професії відповідно до власних здібностей та інтересів з урахуванням потреб суспільства в кадрах. Колективом кафедри гістології, цитології та ембріології були створені оптимальні умови для формування в учнівської молоді розуміння важливості і відповідальності здійснення першого кроку до вибору майбутньої професії, яка б відповідала інтересам, схильностям та здібностям особистості. Старшокласники змогли відчути себе на місці студентів-медиків. Вони отримали змістовну та корисну інформацію щодо особливостей форм навчання на кафедрі, вимог, які висуваються перед здобувачем вищої медичної освіти. Увага школярів була акцентована на важливості опанування такої фундаментальної дисципліни, як гістологія, яка закладає фундамент професійних знань та навичок майбутніх лікарів. Навчання буде цікавим та корисним у разі поєднання його з науково-дослідницькою діяльністю. Сучасна профорієнтаційна робота кафедри гістології, цитології та ембріології є складовою частиною цілісної системи освітнього процесу та спрямована на підготовку молоді до свідомого вибору професії, до самостійного життя, здатності до професійного самовдосконалення, створення сприятливої атмосфери для заохочення до співпраці та активності в трудовому житті.

Аннотация: Макрофаги відіграють центральну роль у тканинному гомеостазі та запаленні. Розуміння походження, розвитку та участі макрофагів, що є резидентними для тканин, у регуляції гомеостатичних процесів є основоположним для розробки майбутніх стратегій втручання для модуляції функцій макрофагів у певних ділянках. Резидентні макрофаги печінки включають клітини Купфера, які разом із синусоїдними клітинами та клітинами Іто утворюють значну популяцію непаренхімних, антигенпрезентуючих клітин. Взаємодія андроген/андрогенний рецептор відіграє роль у розвитку печінки в ембріональній стадії, максимальний диморфізм ефектів настає після статевого дозрівання за рахунок діяльності гіпоталамо-гіпофізарно-гонадної осі. Вплив кверцетину на запальний процес пояснюється блокадою ліпоксигеназного шляху метаболізму арахідонової кислоти, зниженням синтезу лейкотрієнів. Метою нашого дослідження було визначення якісних та кількісних змін у клітинах Купфера при хімічній кастрації самців щурів центрального походження, яка була спричинена введенням розчину триптореліну ацетату. А також визначення результатів дії кверцетину на антигенпрезентуючі клітини печінки на тлі ефектів, викликаних використанням розчину триптореліну. Експерименти проводили на 30 дорослих самцях білих щурів. Щурів розділили на 3 групи: контрольну (10), дослідну 1 (10) та дослідну 2 (10). Тваринам 1 дослідної групи вводили підшкірно триптореліну ацетат у дозі 0,3 мг діючої речовини на кг маси тіла. Тварини 2 дослідної групи отримували триптореліну ацетат у такому ж дозуванні та кверцетин 100 мг на кг маси тіла 3 рази на тиждень. Відмічені зміни в печінці тварин 1 групи на клітинному та субклітинному рівнях, а також у гемомікроциркуляторних судинах, що призвели до незначного венозного застою, сладжу еритроцитів та інфільтрації макрофагами. Одночасне пероральне застосування кверцетину мінімізує структурні та морфологічні зміни в тканині печінки щурів від окисного пошкодження, спричиненого ін’єкцією триптореліну, за рахунок підвищення антиоксидантного захисту тканин печінки.

Аннотация: Розвиток запального процесу в печінці, в тому числі під впливом вірусів гепатиту В і С, контролюється клітинами імунної системи, а саме синусоїдальними ендотеліальними клітинами, клітинами Іто та клітинами Купфера. Макрофаги відіграють одну з ключових ролей у створенні лінії імунного захисту. Яким саме чином певні популяції макрофагів сприяють захворюванням печінки та її регенерації, є предметом постійних дискусій. Тому виявлення особливостей цих популяцій макрофагів людини має беззаперечну цінність при вивченні їх ролі у розвитку патології печінки. Флавоноїд кверцетин має капіляростабілізуючі властивості завдяки антиоксидантному, мембраностабілізуючому ефекту. Метою нашого дослідження було визначити якісні та кількісні зміни імунокомпетентних клітин печінки, спричинені пригніченням центрального синтезу тестостерону у самців щурів внаслідок введення триптореліну ацетату на 270-ту добу, а також потенційний вплив кверцетину на зміну морфології та кількості антигенпрезентуючих клітин печінки на тлі попереднього введення розчину триптореліну ацетату. Експерименти були проведені на 30 статевозрілих самцях білих щурів. Щурів розподіляли на 3 групи: контрольну (10), експериментальну І (ЕГ І, 10) та експериментальну ІІ (ЕГ ІІ, 10). Тваринам з експериментальної групи 1 підшкірно вводили триптореліну ацетат у дозі 0,3 мг активної речовини на кг маси тіла. У експериментальній групі 2 тварини отримували триптореліну ацетат у такому ж дозуванні та кверцетин 100мг на кг маси тіла 3 рази на тиждень, у той час як контрольній групі вводили фізіологічний розчин. Біохімічні дослідження проводили в 10% гомогенаті тканин печінки. Було визначено основна продукція супероксид-аніонного радикала (SAR), а також активність супероксиддисмутази. Пероксинітрит і супероксид аніон-радикал є потужними окислювачами, які можуть пошкоджувати біологічні полімери (ДНК, білки та біологічні мембрани) і призводити до розвитку окисно-нітрозативного стресу.

Аннотация: Метою роботи було вивчення реакції компонентів трійчастого вузла щурів на асептичне запалення в умовах експерименту. Дослідження було проведено на 40 статевозрілих білих щурах-самцях. Об'єктом дослідження був трійчастий ганглій щурів. Тварини були розділені на 2 групи: контрольна група тварин (5), яким вводили ізотонічний розчин NaCl та група експериментальних тварин (35), яким було змодельовано гостре карагененове запалення. Щури були виведені з експерименту на 3, 5, 7, 10, 14, 21, 30 добу шляхом передозування тіопенталового наркозу. Гістологічне дослідження препаратів показало, що карагенен-індуковане запалення в трійчастому ганглії щурів мало стадійний характер перебігу. Реактивні зміни в судинах гемомікроциркуляторного русла були найбільш виражені на 5 добу спостереження, після чого відбувалася поступова стабілізація кровотоку до його повної нормалізації на 14 добу. Ознаки реактивних змін у нейронах у вигляді поступового набухання цитоплазми та ядра перинуклеарного хроматолізу були помітні вже в ранні терміни експерименту. Зі збільшенням термінів спостереження посилювався характер змін. У частини клітин спостерігалися ознаки дегенеративно-дистрофічних процесів. У відповідь зміни нейронів збільшилася проліферативна активність клітин глії. Максимального прояву ознаки запалення в паренхімі досягли на 14-ту добу із незавершеним процесом регенерації до 30-ї доби.

Аннотация: Leydig cells are responsible for the production of testosterone in the male testicles when stimulated by luteinizing hormone (LH). Dysfunction of Leydig cells, which occurs during inflammation or oxidative stress, is one of the main causes of male infertility. The purpose of the study is to determine the effect of quercetin on the microscopic organization of rat testes, nitric oxide production and the intensity of oxidative stress in rat testes on the 90th day of the experiment, during the experimental central deprivation of LH synthesis caused by the administration of triptorelin acetate solution. The experiment was conducted on 20 sexually mature male white rats. Rats were divided into 2 groups of 10 animals in each group: control group (I), group with central deprivation of LH + quercetin synthesis (II). Animals from the group with central blockade of LH synthesis were injected subcutaneously with triptorelin acetate at a dose of 0.3 mg of the active substance per kg and quercetin at 100 mg per kg of body weight 3 times a week, while the control group was injected with saline. Our study of the interstitial space in the testes of white rats showed heterogeneity of macrophage populations and variability of structural and functional parameters. Central blockade of LH synthesis by the administration of treptorelin with the parallel administration of quercitin to the studied animals on the 90th day of the experiment causes changes in the structure of the interstitial space of rat testes, which is characterized by high variability both in the populations of interstitial endocrinocytes and macrophages. Biochemical indicators on the 90th day of the experiment indicate an increase in NO production in conditions of central blocking of the synthesis of luteinizing hormone by more than three times, which is ensured by the activity of the inducible isoform of NOS (iNOS). At the same time, the increase in the activity of iNOS with a decrease in the activity of the arginase pathway leads to the polarization of macrophages according to the pro-inflammatory type. The introduction of quercetin protects the testicular tissue of rats from oxidative damage caused by the administration of triptorelin on the 90th day of the experiment by increasing antioxidant protection and reducing reactive oxygen species in the tissue
Клітини Лейдіга відповідають за вироблення тестостерону в чоловічих яєчках при стимуляції лютеїнізуючим гормоном (ЛГ). Дисфункція клітин Лейдіга, що виникає при запаленнях або окисних стресах, є однією з основних причин чоловічого безпліддя. Мета дослідження – встановити вплив кверцетину на мікроскопічну організацію сім’яників щурів, продукцію оксиду азоту та інтенсивність оксидативного стресу в сім’яниках щурів на 90-ту добу експерименту під час експериментальної центральної депривації синтезу ЛГ, викликаного введенням розчину триптореліна ацетату. Експеримент проводили на 20 статевозрілих самцях білих щурів. Щури були розподілені на 2 групи по 10 тварин у кожній групі: контрольна група (І), група з центральною депривацією синтезу ЛГ + кверцетин (ІІ). Тваринам із групи з центральною блокадою синтезу ЛГ підшкірно вводили триптореліну ацетат у дозі 0,3 мг діючої речовини на кг та кверцетин 100 мг на кг маси тіла 3 рази на тиждень, при цьому контрольній групі вводили фізіологічний розчин. Проведене нами дослідження інтерстиційного простору в сім’яниках білих щурів показало неоднорідність популяцій макрофагів та мінливість структурно-функціональних параметрів. Центральна блокада синтезу ЛГ шляхом введення триптореліну з паралельним введенням кверцитину досліджуваним тваринам на 90-й день експерименту викликає зміни в структурі інтерстиціального простору сім’яників щурів, що характеризується високою варіабельністю як у популяціях інтерстиціальних ендокриноцитів, так і в макрофагів. Біохімічні показники на 90-й день експерименту вказують на підвищення продукції NO в умовах центрального блокування синтезу лютеїнізуючого гормону понад ніж у три рази, що забезпечується активністю індуцибельної ізоформи NOS (іNOS). У той же час підвищення активності іNOS при зниженні активності аргіназного шляху призводить до полярізаціїї макрофагів за прозапальним типом. Введення кверцетину захищає тканину яєчок щурів від окисного пошкодження, викликаного введенням триптореліну, на 90-й день експерименту за рахунок підвищення антиоксидантного захисту та зниження активних форм кисню в тканині

Аннотация: Triptorelin is a gonadotropin-releasing hormone agonist that is a potent inhibitor of testosterone (in men) and estrogen (in women) synthesis and is used to treat advanced prostate cancer. Studies of the mechanisms of regulation and synthesis of testosterone formation in testicular interstitial cells demonstrate multiple endogenous targets that can increase testosterone biosynthesis, which may moderate the effects of testosterone depletion. Triptorelin, a synthetic analog of the neurohormone gonadoliberin, suppresses the expression of the GnRH receptor in the pituitary gland, but does not change the functioning of the pituitary-testicular complex. The purpose of the work is to study the electron microscopic changes in the interstitial endocrinocytes of the testes of rats after the administration of triptorelin for 365 days. The experiment was conducted on 35 sexually mature male white rats. The rats were divided into 2 groups: the control group (I) was injected with a physiological solution, the II group with central deprivation of the synthesis of luteinizing hormone was injected subcutaneously with triptorelin at a dose of 0.3 mg of the active substance per kg of the rat's body weight. The study of the interstitial space in the testicles of white rats showed that long-term administration of triptorelin causes hormonal dysregulation of the hypothalamus-pituitary-testis system, which leads to quantitative and qualitative changes in the endocrine cells of the interstitial space of the testis, which is confirmed by electron microscopic changes in subcellular structures. The maximum effect of triptorelin is determined from the 180th day of observation, which is characterized by an increase in degenerative changes in endocrinocytes, and the detection of Reinke crystals in the cytoplasm of interstitial endocrinocytes from the 270th day of observation
Трипторелін є агоністом гонадотропін-рилізинг-гормону, який є потужним інгібітором синтезу тестостерону (у чоловіків) та естрогену (у жінок) і використовується для лікування прогресуючого раку простати. Дослідження механізмів регуляції та синтезу утворення тестостерону в тестикулярних інтерстиційних клітинах демонструють численні ендогенні мішені, які можуть збільшити біосинтез тестостерону, що може пом’якшити ефекти зниження тестостерону. Трипторелін, синтетичний аналог нейрогормону гонадоліберину, пригнічує експресію рецептора GnRH в гіпофізі, але не змінює функціонування гіпофізарно-тестикулярного комплексу. Мета дослідження – вивчити електронномікроскопічні зміни інтерстиціальних ендокриноцитів сім’яників щурів після введення триптореліну протягом 365 діб. Експеримент проведено на 35 статевозрілих самцях білих щурів. Щури були поділені на 2 групи: контрольна група (І) – вводили фізіологічний розчин, ІІ група з центральною депривацією синтезу лютеїнізуючого гормону – підшкірно вводили трипторелін у дозі 0,3 мг діючої речовини на кг маси тіла щура. Проведене дослідження інтерстиційного простору в сім’яниках білих щурів показало, що довготривале введення триптореліну викликає гормональну дисрегуляцію за системою гіпоталамус-гіпофіз-яєчко, що призводить до кількісних та якісних змін в ендокринних клітинах інтерстиційного простору яєчка, яке підтверджується електронномікроскопічними змінами субклітинних структур. Максимальний вплив триптореліну визначається з 180-ї доби спостереження, що характеризується підвищенням дегенеративних змін в ендокриноцитах та виявленням з 270-ї доби спостереження кристалів Рейнке в цитоплазмі інтерстиційних ендокриноцитів

Аннотация: Малі слинні залози дуже чутливі до патологічних процесів в організмі, однак до теперішнього часу реактивність малих слинних залоз у відповідь на нейроендокринні зміни в організмі вивчена не достатньо і тому є однією з невирішених проблем сучасної морфології. Введення триптореліну негативно впливає на структурні компоненти секреторних та вивідних протоків малих слинних залоз щурів, які проявились статистично достовірним збільшенням всіх структурних компонентів з максимумом на 30-у добу, без відновлення іх до показників контрольної групи на 180-у. Порівнюючи морфометричні показники експериментальної групи на 30-у та 180-у доби визначили статистично достовірне зменшення морфометричних показників, а саме: висота мукоцитів зменшилась на 21 %, зовнішній діаметр кінцевого секреторного відділу на 22 %, просвіт кінцевого секреторного відділу на 27 %; висота головних клітин вивідних проток на 9 %, зовнішній діаметр вивідних проток на 24 %, просвіт вивідних проток на 27 %, на нашу думку, за рахунок зменшення компенсаторних властивостей тканин малих слинних залоз.
